Supplementary Information:

The General Receptor for Phosphoinositides 1 (GRASP) also known as GRP1 is a protein that functions as a phosphoinositide-binding protein and regulates cellular membrane dynamics. It has a mass of approximately 97 kDa. GRASP is expressed in various tissues with significant presence in the brain and immune cells. Mechanically GRASP interacts with phosphoinositides on cellular membranes which assists in membrane trafficking and signaling.
Biological function summary
GRASP has a role in modulating intracellular transport processes and is critical in endosomal trafficking. It operates as part of a larger complex that regulates the recycling of membranes and proteins within cells. GRASP aids in ensuring that the proteins and membranes reach their intended cellular destinations maintaining cellular homeostasis.
Pathways
The protein is an important participant in the phosphoinositide 3-kinase (PI3K) signaling pathway an important pathway in cell growth proliferation and survival. GRASP interacts with proteins such as ARF6 influencing membrane dynamics and cellular signaling. Involved in these pathways GRASP contributes to proper cellular responses to extracellular signals and impacts various downstream biological effects.
GRASP plays a critical role in neurological disorders and immune system irregularities. Disruptions in GRASP function can be associated with conditions like Parkinson's disease and systemic lupus erythematosus (SLE). In Parkinson's GRASP interacts with proteins like LRRK2 suggesting a link between phosphoinositide signaling and neurodegenerative pathogenesis. In the immune context alterations in GRASP function seem connected to immune cell signaling irregularities influencing SLE development.
