The GUCY1A2 protein also known as guanylate cyclase soluble subunit alpha-2 is an enzyme important in the conversion of GTP to cyclic GMP. This protein has a molecular mass of about 77 kDa. GUCY1A2 is expressed in various tissues including the brain heart and lung supporting diverse physiological functions. The enzyme functions as part of a larger heterodimeric complex binding with another subunit to become active.
Biological function summary
This enzyme plays an essential role in the nitric oxide (NO) signaling pathway. GUCY1A2 as a subunit of soluble guanylate cyclase (sGC) complex binds with nitric oxide to produce cyclic GMP a secondary messenger that influences many physiological processes. These processes include vascular smooth muscle relaxation and neurotransmission both of which depend on the synthesis of cyclic GMP. The balance and regulation of this pathway are critical to maintaining normal cell functions.

GUCY1A2 engages significantly in the NO-cGMP signaling pathway and the vascular smooth muscle contraction pathway. Through its role in the NO-cGMP signaling pathway GUCY1A2 collaborates with proteins like sGC subunit beta-1 (GUCY1B1) to facilitate cyclic GMP production. This pathway plays an important function in regulating vascular tone and neuronal signaling both of which are important for homeostasis.
Dysregulation of GUCY1A2 function associates with conditions like pulmonary hypertension and cardiovascular diseases. In pulmonary hypertension the improper production or response to NO impairs normal cGMP synthesis contributing to increased vascular resistance. The protein's interaction with GUCY1B1 becomes relevant here as alterations in GUCY1A2 impact the heterodimeric guanylate cyclase's activity leading to heightened disease risk. Understanding this relationship is key to developing targeted therapeutics for related disorders.
