PROKR2 also known as the Prokineticin Receptor 2 is a G protein-coupled receptor with a molecular weight of approximately 43 kDa. It binds to prokineticins which are small proteins involved in various biological processes. PROKR2 is expressed prominently in the central nervous system particularly in the hypothalamus and also in peripheral tissues such as the testis and the pituitary gland. The receptor when activated by its ligand facilitates intracellular signaling through downstream effectors.
Biological function summary
This receptor plays an important role in the regulation of circadian rhythm and reproductive functions. PROKR2 forms part of a signaling complex that influences the release of gonadotropin-releasing hormone. It partners with prokineticin ligands to mediate its effects orchestrating processes like sleep-wake cycles and olfactory bulb function. Its activity is important in maintaining normal physiological functions related to these processes.
Pathways
PROKR2 is deeply integrated into the hypothalamic-pituitary-gonadal (HPG) axis pathway influencing reproductive hormone release. Another important pathway is the circadian rhythm pathway where it links with proteins such as CLOCK and BMAL1. These pathways are essential for the daily regulation of biological rhythms and reproductive health aligning with environmental cues like light and darkness.
PROKR2 has associations with Kallmann syndrome a condition characterized by anosmia and hypogonadotropic hypogonadism. The receptor's abnormal expression or function can lead to impaired gonadal development. Another disorder related to PROKR2 is obesity where its signaling pathways intersect with leptin’s activity affecting body weight regulation. Understanding these connections is essential for developing therapeutic interventions in related disorders.
