The PML-RARA fusion protein also known as PML-RARA or PML/RARA is a unique product of a chromosomal translocation between the promyelocytic leukemia (PML) gene on chromosome 15 and the retinoic acid receptor alpha (RARA) gene on chromosome 17. This fusion results in a chimeric protein that typically weighs around 100 kDa. The PML-RARA protein is expressed in the nucleus of cells where it plays an important role in altering gene expression and disrupting normal cellular function.
Biological function summary
The fusion protein impacts transcriptional regulation and is part of a repressive complex. The PML portion of the fusion displaces normal PML protein functions disrupting nuclear body formation while the RARA portion allows it to bind to DNA and recruit co-repressors affecting gene expression. It inhibits granulocyte differentiation an essential process for the maturation of blood cells leading to the accumulation of immature promyelocytes.
Pathways
The PML-RARA protein interferes with the retinoic acid signaling pathway affecting the balance of cellular growth and differentiation. It interacts with proteins such as RXR (Retinoid X Receptor) which helps regulate transcription of genes involved in cell differentiation. PML-RARA also impacts the response to vitamin A metabolites making it an important player in altering pathways critical to hematopoiesis.
The PML-RARA fusion protein is a hallmark of acute promyelocytic leukemia (APL) a subtype of acute myeloid leukemia (AML). The presence of this fusion protein contributes to the pathogenesis of APL by blocking differentiation of myeloid lineage cells. RARA is specifically involved in this disease process and treatments targeting PML-RARA such as all-trans retinoic acid (ATRA) leverage its effect on this fusion protein to induce differentiation and remission in affected individuals.
