The ASC2/ASCI protein also known as nuclear receptor coactivator 7 (NCOA7) is an intracellular protein with a mass of approximately 87 kDa. It is mainly expressed in the brain pancreas and testis indicating its significant roles in these tissues. ASC2 acts as a transcriptional coactivator by enhancing the transcriptional activities of steroid hormone receptors and nuclear receptors. Through its coactivator functions ASC2 interacts with other proteins in the nucleus to help regulate the transcription of specific genes.
Biological function summary
ASC2 contributes as a part of the larger coactivator complex that enhances gene transcription. It interacts with nuclear receptors to potentiate their ability to bind DNA and influence gene expression. This protein also engages in direct protein-protein interactions with other transcription factors such as ASCOM components facilitating a range of cellular functions. ASC2's presence in multiple tissues suggests its involvement in diverse biological processes including cellular growth and differentiation.
Pathways
ASC2 plays a role in important signal transduction pathways such as the nuclear receptor signaling pathway. It works alongside proteins like estrogen receptor and androgen receptor which are integral to the regulation of genes involved in cellular homeostasis and metabolism. ASC2 as part of the larger transcriptional machinery assists in fine-tuning the expression of genes in response to hormonal signals and environmental cues.
ASC2 has been linked to neurodegenerative diseases like Alzheimer's disease and certain types of cancer. In Alzheimer's disease the interaction of ASC2 with estrogen receptors can influence amyloid beta clearance affecting disease progression. In cancers ASC2’s ability to amplify nuclear receptor activities suggests its involvement in tumor growth dynamics. Its interactions with coactivators in these disorders make ASC2 a target of interest for therapeutic intervention.
