PTP4A3 also known as PRL-R is a protein tyrosine phosphatase with a molecular mass of approximately 19 kDa. This protein is expressed in various tissues but has high levels in cancerous cells. It has a role in the dephosphorylation of substrates impacting cell processes like proliferation and motility. PTP4A3 localizes to the cytoplasm and its enzymatic activity can influence cellular dynamics particularly in rapidly dividing cells.
Biological function summary
PTP4A3 affects cellular growth and division as part of the signaling network. It is involved in the intracellular control of the phosphatase complex and regulates other protein interactions. PTP4A3 influences cell migration and invasion particularly through interactions with structural proteins. As a phosphatase it removes phosphate groups influencing the phosphorylation status of other proteins and altering their functions in the signaling pathways.
Pathways
PTP4A3 takes part in processes like the MAPK and PI3K/AKT signaling pathways. These pathways are critical for cell survival proliferation and differentiation. Within these pathways PTP4A3 modulates interactions with proteins like ERK1/2 and AKT which are important components of the pathways. The phosphatase activity facilitates crosstalk and integration of signals maintaining cellular homeostasis.
Aberrant PTP4A3 expression connects to cancer particularly in metastatic progression. Elevated levels of PTP4A3 correlate with aggressive tumor behavior and poor prognosis. In addition PTP4A3 affects chronic inflammatory disorders through its actions on signaling pathways. It impacts epithelial to mesenchymal transition (EMT) processes linking to proteins like MMPs which play a role in cancer metastasis and inflammation.
