Alpha-synuclein phosphorylated at serine 129 (pS129; phosphorylated alpha synuclein, SNCA/NACP) is a modified form of the alpha-synuclein protein, which plays a crucial role in the regulation of synaptic vesicle trafficking and neurotransmitter release. This phosphorylation is significantly increased in the brains of patients with Parkinson's disease and other synucleinopathies. The presence of pS129 is associated with the formation of Lewy bodies, a hallmark of Parkinson's disease, and is believed to contribute to the neurotoxic properties of alpha-synuclein, exacerbating disease progression.

Alpha-synuclein also known as SNCA or PARK1 protein is a monomer protein with a molecular weight of approximately 14 kDa. It plays an important role in the synaptic functions of neurons. You usually find this protein expressed in the brain specifically in the presynaptic terminals of neurons. It also appears in other tissues including the heart and muscles though to a lesser degree. In its natural state alpha-synuclein exists as a soluble monomer but under certain conditions it can form insoluble protein filaments. The antibodies 4d6 and ep1536y might be used to study this protein providing tools to investigate its role and behavior in biological processes.
Biological function summary
Alpha-synuclein contributes to the regulation of synaptic vesicle trafficking and release. It interacts with the cell membrane and is involved in the assembly of SNARE complexes important for neurotransmitter release. Although alpha-synuclein generally acts in its monomeric form it may also aggregate and become involved in the assembly of fibrils under pathological conditions impacting normal cellular function. The protein's ability to interact with lipid membranes suggests its possible role in maintaining membrane integrity and dynamics.

Alpha-synuclein is integral to neurotransmitter release and synaptic plasticity influencing dopaminergic signaling pathways. This protein interacts with other synaptic proteins like synaptophysin and synaptotagmin linking it to vesicular transport and recycling pathways. Alpha-synuclein is strongly associated with neurodegenerative diseases like Parkinson's disease and Lewy body dementia.
