Activated Notch1 also known as cleaved Notch1 and Notch 1 NICD is an intracellular domain resulting from Notch1 receptor cleavage. The molecular weight of Notch1 NICD varies but it is generally around 120-150 kDa depending on post-translational modifications. These receptors are present in many cell types including embryonic cells stem cells and various tissue-specific cells. They reside in the cell membrane and become activated upon interaction with specific ligands.
Biological function summary
Activated Notch1 is critical for regulating cell fate decisions including proliferation differentiation and apoptosis. It participates in a multi-protein complex that includes the CSL (CBF1/Su(H)/LAG-1) transcription factor modulating gene expression by binding to DNA. The role of activated Notch1 in maintaining the balance between cell growth and death is important in tissue homeostasis and development.
Pathways
Activated Notch1 plays a central role in the Notch signaling pathway an essential pathway for intercellular communication. This pathway involves several proteins including Delta and Jagged as ligands. It contributes to several critical developmental processes and is connected with the regulation of the NF-kB signaling pathway involved in immune responses and inflammation.
Activated Notch1 is significantly associated with T-cell acute lymphoblastic leukemia (T-ALL) and breast cancer. Abnormal expressions or mutations in Notch1 can lead to unchecked cellular proliferation in these conditions. Notch1 alterations often coexist with disruptions in other signaling proteins such as p53 contributing to disease pathology and presenting potential targets for therapeutic intervention.
