The Cdk4 + Cyclin D1 complex plays an important role in cell cycle regulation specifically during the G1 phase. Cdk4 also known as cyclin-dependent kinase 4 has a molecular weight of approximately 34 kDa. This protein is expressed in various tissues particularly active in cells undergoing proliferation. Cyclin D1 often referred to simply as cyclin D partners with Cdk4 to form a functional complex. The Cyclin D1 + Cdk4 protein interaction is essential for the phosphorylation of the retinoblastoma protein (Rb) allowing progression through the cell cycle.
Biological function summary
Cyclin D1 and Cdk4 are important for progression of the cell cycle from G1 to S phase. The Cyclin D1 + Cdk4 protein complex acts by phosphorylating Rb releasing E2F transcription factors which initiate DNA replication. This complex therefore serves as a regulatory node controlling cell cycle entry and is influenced by mitogenic signals. Its activity is tightly regulated in response to extracellular signals aligning cell cycle progress with growth conditions making it a pivotal player in cell division.

They are integral to the MAPK/ERK pathway which links external growth signals to cell cycle entry and the PI3K/AKT pathway which provides additional control over cell survival and metabolism. Through these pathways Cyclin D1 and Cdk4 interact with other proteins like p21 and p27 which modulate their activity and ensure proper cell cycle transitions.
The Cyclin D1 + Cdk4 complex has significant implications for oncogenesis including breast cancer and melanoma. Overexpression or dysregulation of Cyclin D1 or Cdk4 can lead to unchecked cell proliferation due to faulty cell cycle control. In breast cancer Cyclin D1 overexpression often correlates with ER-positive tumors while in melanoma Cdk4 mutations are associated with resistance to conventional therapies. Proteins such as p16INK4a an inhibitor of Cdk4 activity also relate to these cancers as their functional loss can contribute to disease progression.
