Trichohyalin (THH) acts mechanically as a structural protein that plays an important role in hair and skin cell cornification. Expressed in the inner root sheath of hair follicles and in the granular layer of the epidermis it contributes to cell envelope formation. This protein with a molecular mass of approximately 250 kDa is also known as trichohyalin-1. THH is rich in glutamine and lysine which allows it to bind to other proteins and form a stable matrix structure in hair and skin.
Biological function summary
Trichohyalin contributes to the development and maintenance of hair follicle integrity and skin barrier function. As part of the keratinocyte differentiation process it interacts with various keratins and contributes to cell envelope formation by reinforcing cell membranes through cross-linking. The protein forms a part of the cornified cell envelope complex where it binds with filaggrin and loricrin aiding in cellular structural support and resistance to mechanical stress.
