HePTP also known as Hematopoietic Protein Tyrosine Phosphatase or PTPN7 is a non-receptor protein tyrosine phosphatase with a molecular mass of approximately 37 kDa. It possesses a catalytic domain which is typical for PTP family members. HePTP dephosphorylates phosphotyrosine residues acting as a regulatory switch in signaling pathways. This protein is expressed mainly in hematopoietic tissues such as thymus spleen and peripheral blood leukocytes indicating its role in immune function.
Biological function summary
HePTP affects T cell activation and function by modulating the phosphorylation state of target proteins. It is involved in forming complexes with MAP kinases such as ERK and p38. Phosphorylation changes in these target proteins influence cellular responses like differentiation proliferation and apoptosis emphasizing the regulatory importance of HePTP in immune responses.
Pathways
HePTP participates in the MAPK signaling pathway where it inhibits MAP kinase activity by dephosphorylation. This action influences downstream processes that control cell cycle apoptosis and cellular stress responses. HePTP interacts with proteins such as ERK2 and is functionally related to other phosphatases like DUSP3 playing a critical role in the balance of phosphorylation within the cell signaling network.
HePTP links to immune-related conditions such as rheumatoid arthritis and allergies. Its activity modulation may impact the signaling pathways involved in these diseases potentially affecting disease progression and treatment. HePTP associates with proteins such as SHP-2 in rheumatoid arthritis contributing to the complex pathophysiological mechanisms underlying this disorder.
