The GRIK2 protein also known as GluK2 is a subunit of the kainate receptor which is part of the ionotropic glutamate receptor family. This receptor primarily mediates excitatory synaptic transmission in the central nervous system. GRIK2 has a molecular weight of approximately 104 kDa and is highly expressed in regions such as the hippocampus cortex and cerebellum. These areas are critical for learning and memory processes which underlines the importance of GluK2 in neuronal communication.
Biological function summary
GRIK2 plays a significant role in modulating synaptic plasticity an important mechanism for learning and memory. It forms heteromeric complexes with other kainate receptor subunits like GluK1 and GluK3 which can affect receptor properties. This combination allows for diverse functional responses and adaptability in synaptic transmission. The presence of GRIK2 in neurons indicates that it is integral in processes such as synaptic strength modulation and response to different synaptic inputs.
Pathways
GRIK2 is essential in the glutamatergic signaling pathway which is involved in many excitatory transmissions within the brain. This protein interacts with associated proteins such as PSD-95 and PICK1 which helps in receptor clustering and synaptic signaling regulation. Additionally GRIK2 is involved in calcium signaling pathways through its ability to regulate calcium influx contributing to neuron function and communication.
GRIK2 has been linked to conditions such as epilepsy and schizophrenia. Mutations or dysregulation in the expression of GRIK2 can affect neurotransmission balance which can lead to these neurological disorders. Its interaction with other receptor subunits like GluK1 and GluK3 also connects GRIK2 to these diseases since alterations in the complex can disrupt normal synaptic function. Understanding these relationships is key in developing therapeutic strategies for such disorders.
