The HIV-1 Vif protein also known as Virion Infectivity Factor plays an essential mechanical role in the life cycle of the HIV virus. Expressed in infected host cells Vif is a 23 kDa protein that facilitates viral replication by counteracting the host's intrinsic antiviral defenses. Vif complexes with cellular proteins to achieve this function and is an important component of the viral assembly process playing an important part in ensuring virus infectivity.
Biological function summary
Vif disrupts host antiviral mechanisms by targeting the host enzyme APOBEC3G a cytidine deaminase for proteasomal degradation. By doing so Vif prevents the incorporation of APOBEC3G into new virions which would otherwise inhibit reverse transcription in the next round of infection. Vif forms an E3 ubiquitin ligase complex that includes several host proteins which is important for initiating the ubiquitination process leading to APOBEC3G degradation.
Pathways
Vif is centrally involved in the HIV-1 lifecycle pathway. It ensures the success of HIV replication by neutralizing antiviral activity before the incorporation of APOBEC3G into virions. Through these pathways Vif interacts with the cellular ubiquitin-proteasome system and engages with proteins like CBF-beta and Cul5 from the ubiquitin ligase complex illustrating Vif's role in manipulating host cellular machinery to benefit viral propagation.
HIV-1 Vif is intimately connected to the progression of HIV/AIDS. By disabling APOBEC3G Vif promotes the persistence of HIV in the host allowing the virus to evade innate immune responses. The failure of the immune system in controlling the virus leads to the gradual development of AIDS. Vif's actions are closely linked to CD4 T-cell depletion a hallmark of AIDS and highlight the critical role of Vif in linking molecular interactions with disease progression primarily through its interaction with APOBEC3G.
