Leupaxin is a cytoskeletal-associated protein involved in cellular adhesion and migration. Also known as LPXN it belongs to the paxillin superfamily and has a molecular weight of approximately 42 kDa. The expression of leupaxin is mainly found in hematopoietic cells including leukocytes. It contains LIM domains and an LD motif which play significant roles in protein-protein interactions.
Biological function summary
The protein plays an important role in regulating actin cytoskeleton dynamics and cell spreading. It interacts with various signaling complexes and integrates into focal adhesions. Leupaxin often participates with kinases such as FAK and Src to modulate cellular responses to external stimuli. Its colocalization with focal adhesion proteins suggests involvement in mechanotransduction processes.
Pathways
Proteins like leupaxin participate in pathways regulating cell migration and adhesion. One notable pathway is the integrin signaling pathway influencing diverse cellular processes. It associates with proteins like talin and vinculin essential for the linkage between integrins and the cytoskeleton. These interactions help in modifying cytoskeletal reorganization and cellular signaling cascades.
Leupaxin has relevance in cancer particularly in metastatic processes and autoimmune disorders. Its role in promoting or inhibiting cell migration ties into metastatic cancer's progression where it possibly interacts with proteins like paxillin. In autoimmune disorders aberrant function or expression of leupaxin could affect immune cell mobility and contribute to disease pathology by modifying immune cell adhesion properties.
