IMPAD1 also known as Inositol Monophosphatase Domain Containing 1 is an enzyme with a molecular mass of approximately 50 kDa. The enzyme is primarily located in the Golgi apparatus of cells. It catalyzes the dephosphorylation of inositol monophosphates which is a critical step in the phosphatidylinositol signaling pathway. The expression of IMPAD1 occurs in various tissues including the brain cartilage and kidney reflecting its diverse roles in different biological functions.
Biological function summary
IMPAD1 contributes to maintaining cellular inositol balance and is involved in the regulation of signaling pathways. It does not directly participate in a complex but its activity is essential for sustaining the levels of inositol phosphates within the cell. This function is particularly significant in neural and cartilage cells where it helps in the modulation of cellular responses to external stimuli.
Pathways
IMPAD1 plays a role in the phosphoinositide signaling pathway and the inositol phosphate metabolism pathway. It works alongside other phosphatases and kinases including Inositol polyphosphate-4-phosphatase type II (INPP4B) to regulate the phosphorylation state of inositol phosphates. Through these pathways it influences the synthesis of important secondary messengers like PI(345)P3 which are important for cellular signaling processes.
IMPAD1 has been associated with chondrodysplasia with joint dislocations and neurodegenerative disorders. Mutations in the IMPAD1 gene can disrupt the inositol metabolism leading to severe skeletal abnormalities and joint issues. Additionally dysfunction in its related signaling pathways particularly through interactions with proteins like INPP4B can contribute to neurodegenerative pathologies highlighting its broad impact on human health.
