The DHX8 also known as Prp22 is a protein involved in RNA processing. Mechanically it functions as an ATP-dependent RNA helicase. This protein facilitates the unwinding of RNA structures during the spliceosome cycle essential for accurate splicing of pre-mRNA to mature mRNA. The molecular weight of DHX8/Prp22 is approximately 140 kDa. It is widely expressed in animal cells particularly in tissues with high levels of RNA processing activity such as the liver and testis.
Biological function summary
DHX8/Prp22 plays a critical role in pre-mRNA splicing. It is a component of the spliceosome a large ribonucleoprotein complex responsible for excising introns from pre-mRNA. Within this complex DHX8/Prp22 helps in the transition to the second step of splicing by promoting the release of spliced mRNA and dissociation of proteins and snRNPs. The activity of DHX8/Prp22 ensures proper removal of introns and prevents exon skipping contributing to the fidelity of gene expression.
Pathways
The DHX8/Prp22 is an integral part of the cellular RNA splicing pathway. It connects with the spliceosomal cycle and influences mRNA maturation which is essential for proper gene expression and cellular function. This protein has associations with other helicases like DHX15 which also work in mRNA splicing pathways indicating a complex interplay of helicases in gene expression regulation. Splicing impacts various cellular processes linking DHX8/Prp22 to larger networks of gene expression regulation.
DHX8/Prp22 is linked to certain genetic diseases where splicing errors occur such as retinitis pigmentosa and some cancers. Improper functioning of DHX8/Prp22 can lead to aberrant splicing resulting in the production of dysfunctional proteins. This helicase interacts with other splicing factors such as SF3B1 mutations of which are found in these conditions highlighting the importance of spliceosome integrity in disease prevention. Understanding DHX8/Prp22's role in these disorders can lead to potential therapeutic strategies targeting splicing mechanisms.
