CENH3 also known as Centromere Protein A is a histone H3 variant that plays an essential role in the packaging of centromeric DNA. This protein has a mass of approximately 17 kDa and is uniquely expressed in the centromeric chromatin of eukaryotic cells. CENH3 differs from the canonical H3 histone by having a distinct N-terminal tail which is important for its specific incorporation into centromeres. This specific localization is necessary for the recognition and binding of other centromeric proteins facilitating the assembly of the kinetochore complex.
Biological function summary
CENH3 integrates into nucleosomes at the centromeres forming a specialized chromatin structure critical for chromosome segregation. It contributes to the foundation of the centromere by assembling into a larger protein complex known as the kinetochore. This complex mediates the attachment of spindle microtubules during mitosis ensuring accurate distribution of chromosomes to daughter cells. CENH3's presence at the centromeres helps in recruiting other centromeric proteins such as CENP-C and CENP-N highlighting its central role in stabilizing the centromeric region.

The proper function of CENH3 influences pathways connected to Aurora B kinase activity which affects the correction of misattached microtubules ensuring accurate and efficient mitotic progression.
CENH3 abnormalities can contribute to conditions connected to chromosome missegregation and aneuploidy. Misregulation of CENH3 expression or function links to developmental disorders like cancer and congenital disorders. Dysregulated CENH3 may impact proteins such as SGO1 an essential kinetochore protein that directly affects chromosome cohesion and separation. Abnormal CENH3 expression can result in improper kinetochore function leading to an increased risk of chromosomal instability and tumorigenesis.
