MKRN2 also known as Makorin ring finger protein 2 functions mechanically as an E3 ubiquitin ligase. It possesses a molecular mass of approximately 49 kDa. This protein contributes to the ubiquitination process by tagging specific substrate proteins for degradation. MKRN2 is expressed broadly across various tissues with higher levels observed in neural tissues. This suggests its role in neural functions although it is not exclusive to the nerves.
Biological function summary
MKRN2 participates in numerous cellular functions through its role in ubiquitination. Its presence in neural tissues indicates a significant involvement in neurodevelopmental processes and neural signaling. MKRN2 interacts as part of a complex with other proteins that mediate protein stability and degradation. This interaction highlights its multifunctional role in cellular homeostasis suggesting its influence extends beyond simple protein tagging.
Pathways
MKRN2 engages in critical signaling cascades such as the Wnt and PI3K/Akt pathways. Within the Wnt pathway MKRN2 can modulate the stability of proteins involved in signal transmission such as β-catenin. The PI3K/Akt pathway involves MKRN2 in regulation of cell survival and growth often linking it with other proteins like PTEN. These pathways indicate MKRN2’s broader importance in cell proliferation and apoptosis control.
MKRN2 has connections with cancer and neurodegenerative diseases. Mutations or dysregulation of MKRN2 can lead to aberrant protein degradation contributing to tumorigenesis and progression of cancers. The protein also shows associations with neurodegenerative disorders through its interaction with substrates involved in neural functioning. Another protein linked to these disorders includes p53 where MKRN2 can influence cell cycle regulation and stress responses in the neural context.
