APOBEC1 also known as Apolipoprotein B mRNA Editing Enzyme Catalytic Subunit 1 is an enzyme responsible for the deamination of cytidine to uridine in mRNA. This editing capability focuses mainly on apolipoprotein B (apoB) mRNA affecting lipid metabolism significantly. The APOBEC1 protein weighs approximately 27 kDa. It expresses mainly in the small intestine and liver where lipid processing is an essential biological function.
Biological function summary
APOBEC1 plays an important role in lipid homeostasis by editing apoB mRNA transcripts leading to the production of a shorter form of the apoB protein. This protein acts as a critical component in lipid transport and metabolism. APOBEC1 functions within a multisubunit complex that includes other proteins like A1CF (APOBEC1 Complementation Factor). This protein complex ensures efficient mRNA editing necessary for optimal lipid regulation.
Pathways
The APOBEC1 enzyme is part of key metabolic routes notably lipid metabolism and lipid-transport routes. It contributes to the post-transcriptional modification of apoB mRNA influencing pathways like lipoprotein assembly. Through these pathways related proteins such as microsomal triglyceride transfer protein (MTP) work alongside APOBEC1 to facilitate lipid processing. MTP assists in lipoprotein biogenesis highlighting a collaborative function in lipid management.
APOBEC1 has associations with conditions such as atherosclerosis and hepatic steatosis. Aberrant expression or function of APOBEC1 can lead to improper lipid metabolism directly linking to the development of atherosclerosis due to the accumulation of cholesterol-rich lipoproteins. Furthermore hepatic steatosis or fatty liver disease may arise when lipid homeostasis deteriorates. In these conditions APOBEC1 along with proteins like apoB becomes central to understanding disease mechanisms and potential therapeutic interventions.
