SPNS2 also known as sphingosine-1-phosphate transporter 2 is a membrane protein with a mass of approximately 58 kDa. It functions to transport sphingosine-1-phosphate (S1P) a signaling lipid across cell membranes. SPNS2 shows expression in various tissues including the lymphatic endothelium. This transporter plays an important role in regulating extracellular S1P levels influencing the gradient between tissues and circulation.
Biological function summary
SPNS2 facilitates S1P export impacting immune cell trafficking vascular maturation and heart development. It operates as part of a transporter complex influencing the localization and release of S1P to maintain proper signaling and function in different physiological contexts. The presence of SPNS2 affects the distribution and gradient of S1P an important signaling molecule involved in multiple cellular processes.
Pathways
This transporter integrates in S1P signaling pathways and the immune response pathway. SPNS2 modulates the S1P gradient necessary for the migration of lymphocytes engaging in interaction with S1P receptors particularly S1PR1. These interactions help to control lymphocyte egress from lymphoid organs and contribute to vascular integrity and function.
SPNS2 associates with autoimmune diseases and cancer progression. Altered SPNS2 expression can result in dysregulation of the S1P gradient impacting conditions such as multiple sclerosis due to disrupted lymphocyte trafficking. In cancer aberrant S1P signaling in which SPNS2 plays a role can influence tumor growth and metastasis often through interaction with other proteins involved in the S1P pathway like S1PR1.
