ARHGAP29 also known as Rho GTPase Activating Protein 29 is a protein involved in regulating the actin cytoskeleton through its action on Rho family GTPases. This protein has a molecular mass of approximately 145 kDa. It is expressed in various tissues including the heart and the brain. ARHGAP29 acts by stimulating GTPase activity resulting in the inactivation of Rho GTPases which are important for cell morphology migration and adhesion processes.
Biological function summary
ARHGAP29 plays roles in modulating cellular structures and motility by regulating Rho GTPase activity. It functions as a GTPase-activating protein that converts active GTP-bound RhoA Rac1 and Cdc42 to their inactive GDP-bound states. ARHGAP29 is not part of any known major protein complex but interacts with signal transduction molecules to exert its effects on the cytoskeleton.
Pathways
ARHGAP29 is an important regulator within the Rho GTPase signaling pathway influencing actin dynamics that affect cell movement and shape. This protein interacts with and modulates RhoA activity which is involved in the MAPK signaling pathway linking it to broader cellular functions such as growth and differentiation. ARHGAP29 has a relationship with other GTPase-activating proteins collectively contributing to the regulation of the Rho signaling pathway.
ARHGAP29 has been associated with particular conditions like nonsyndromic cleft lip with or without cleft palate and vascular diseases. Mutations or dysregulation of ARHGAP29 can impact cytoskeletal organization influencing tissue morphogenesis and vascular integrity. It connects to other proteins like RhoA whose abnormal regulation often results in similar developmental and vascular disorders.
