FGF5 also known as fibroblast growth factor 5 is a protein involved in various cellular processes with a molecular mass of approximately 29 kDa. It is part of the FGF family which includes at least 22 members playing roles in cell growth differentiation and regulation. FGF5 expression occurs mainly in hair follicles skeletal muscles and the central nervous system. The protein has been identified in epithelial cells and the stromal compartment surrounding hair follicles.
Biological function summary
FGF5 functions as a signaling molecule that influences hair growth cycle regulation. It is an important player in the transition of hair follicles from the anagen (growth) phase to the catagen (regression) phase. FGF5 does not act alone but interacts with various cellular receptors particularly FGFR1 and FGFR2 which facilitate its role in hair cycle modulation. It is not typically part of a stable multi-protein complex but works as a soluble factor influencing nearby cell behavior.
Pathways
FGF5 engages with signaling cascades such as the MAPK/ERK pathway and the PI3K/AKT pathway which are significant for cell proliferation and survival. FGF5 can modulate these pathways by binding to FGFR1 and FGFR2 receptors leading to downstream signaling that affects cell behavior. These pathways illustrate its involvement with several other FGF family members such as FGF7 and FGF10 showcasing a broad effect on cellular dynamics and development.
FGF5 has been linked to conditions such as congenital hypotrichosis and alopecia where abnormalities in hair growth cycles occur. Dysfunctions in FGF5 signaling can lead to prolonged anagen phases or premature catagen transitions. Additionally FGF5 has connections to cancer development due to its role in cell proliferation and differentiation; aberrations in FGF5 activity can contribute to tumorigenesis. Although not directly linked proteins like FGFR1 can mediate these effects increasing the complexity of its involvement in disease progression.
