OBFC1 also known as oligonucleotide/oligosaccharide-binding fold containing one plays a significant role in DNA replication and telomere maintenance. This protein is part of a heterotrimeric complex and has a molecular weight of approximately 55 kDa. The OBFC1 protein is expressed in various tissues with notable expression in the bone marrow where it actively participates in cellular functions. Researchers have studied this protein extensively due to its critical involvement in telomere length regulation and DNA replication processes.
Biological function summary
Oligonucleotide/oligosaccharide-binding fold containing one acts as a part of the CST complex working alongside CTC1 and TEN1. This complex resembles replication protein A (RPA) and mainly functions at telomeres protecting chromosome ends from DNA damage responses. The CST complex competes with the RPA complex and binds to single-stranded DNA to facilitate telomere replication. Moreover OBFC1 has roles in stabilizing telomere structures important in preventing chromosome instability and maintaining genome integrity.

OBFC1 shows involvement in the telomere maintenance pathway which plays an essential role in genome stability and the DNA replication initiation pathway. Through its action in these pathways OBFC1 is closely related to the POT1 protein which binds to telomeric DNA as part of the shelterin complex. Together with other proteins in these pathways OBFC1 helps regulate telomere length and functionality ensuring proper chromosome end protection and replication.
OBFC1 has connections to various conditions involving telomere dysfunction. Two notable disorders include dyskeratosis congenita and some forms of cancer where telomere maintenance mechanisms fail. OBFC1 in association with the protein TERT which encodes the telomerase reverse transcriptase plays a substantial role in the pathogenesis of these diseases by influencing telomere length homeostasis.
