TLE2 also known as Transducin-Like Enhancer of Split 2 is a member of the Groucho/TLE family of co-repressors. It has a molecular mass of approximately 82 kDa. TLE2 is expressed in various tissues including brain and muscle. As a protein it interacts with transcription factors repressing transcription by binding to DNA-binding proteins which ultimately influences gene expression and cellular differentiation.
Biological function summary
TLE2 serves as a general transcriptional co-repressor. It forms complexes with other transcriptional regulators to modulate gene expression participating in the repression of transcriptional activity. TLE2 shows a significant role in embryonic development and neuronal differentiation working alongside other Groucho family members to guide cell fate decisions during development.
Pathways
TLE2 interacts with a range of signaling mechanisms involved in development and differentiation. It is notably involved in the Notch signaling pathway which regulates cell differentiation and proliferation. Additionally TLE2 associates with Wnt signaling interacting with other proteins like LEF1 and TCF to suppress Wnt target genes. These interactions contribute to maintaining a balance between cell proliferation and differentiation.
TLE2 has connections to several neurological conditions. Altered expression or function of TLE2 is linked to neurodevelopmental disorders including autism spectrum disorder. Furthermore disruptions in its interaction with other proteins like LEF1 influence pathological processes associated with cancer particularly in brain tumors. Understanding TLE2's role in these conditions helps in unraveling the molecular mechanisms underlying disease progression.
