Exonuclease 1 (Exo1) also known simply as Exonuclease I is a protein playing a significant mechanical role in DNA processes. The protein functions by degrading nucleic acid molecules from their ends specifically acting on the 5' ends to progressively remove nucleotides. Exo1 exhibits specificity for double-stranded DNA and participates mainly in DNA repair and recombination. The molecular weight of Exo1 is approximately 94 kDa. Expression of Exo1 occurs in various tissues with prevalent expression in tissues undergoing active cell division.
Biological function summary
Exo1 is key in maintaining genomic stability. This protein engages in homologous recombination mismatch repair and the resection process during DNA double-strand break repair. Exo1 works collaboratively as part of larger complexes particularly interacting with proteins like MRE11-RAD50-NBS1 (MRN) complex in DNA repair mechanisms. The interaction within these complexes enhances its efficacy in processing DNA ends and facilitates accurate repair.

Exo1 is integral to the DNA repair and homologous recombination pathways.
