Alpha defensin 1 2 and 3 often called human neutrophil peptides (HNP) 1 2 and 3 are small antimicrobial peptides with masses around 3-4 kDa. You commonly find them in neutrophils and Paneth cells of the small intestine. These peptides display broad-spectrum antimicrobial activity targeting bacteria fungi and some viruses. They insert themselves into microbial membranes causing membrane disruption and cell death. Despite their small size the alpha-defensins have significant effects on microbial populations promoting innate immunity.
Biological function summary
Alpha defensins participate in host defense by exerting antimicrobial action and immunomodulatory functions. They form part of the innate immune system and are integral to the antimicrobial complexes in neutrophils. The peptides interact with other immune components to modulate inflammation and enhance phagocytosis. By recruiting and activating immune cells alpha defensins help manage infections and influence the immune response.
Pathways
Alpha defensins are integral to the immune response and inflammation pathways. They interact with proteins like toll-like receptors (TLRs) to initiate signaling cascades that lead to cytokine production. Additionally the peptides participate in reactive oxygen species (ROS) generation which further aids microbial killing. Their role in these pathways allows immune coordination at the site of infection and contributes to pathogen clearance.
Alterations in alpha defensins' expression or function can link to inflammatory bowel disease (IBD) and cystic fibrosis. In IBD reduced alpha defensin levels can impair mucosal defense leading to increased bacterial translocation and inflammation. The peptides also connect to cystic fibrosis through their role in airway surface immobility and infection management. In these diseases alpha defensins interact with proteins like mucin to influence disease progression and patient outcomes.
