Preparation And Storage: Preparation And Storage The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C.
Recommended Assay Procedures: Recommended Assay Procedures ELISA Capture : The purified BVD8-3G11 antibody (Cat. No. 554672) is useful as a capture antibody in a sandwich ELISA for measuring human IL-3 protein levels. Purified BVD8-3G11 antibody can be paired with the biotinylated BVD3-1F9 antibody (Cat. #554674) as the detecting antibody, with recombinant human IL-3 (Cat. No. 554604) as the standard. This purified capture antibody should be pretitrated in the range of 1 - 4 µg/ml to determine its optimal concentration for ELISA. To obtain linear standard curves, doubling dilutions of human IL-3 ranging from ~2,000 to 15 pg/ml should be used. Note: This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. These ELISA reagents are not recommended for assay of serum samples. WB: The BVD8-3G11 antibody has been found useful for western blotting. A concentration of 1-5 µg/ml (in conjunction with the AKP-conjugated goat anti-rat Igs has been found to enable visualization of ≤ 100 ng/lane of recombinant IL-3 (e.g., Cat. No. 554604), under reducing conditions. Neutralization: The BVD8-3G11 antibody (Cat. No. 554671) is useful for neutralization of human IL-3 bioactivity.
