Mcl-1 is a member of the pro-survival BCL-2 family of apoptotic regulatory proteins. Mcl-1 plays a crucial role in the survival and development of various cell types by binding to proapoptotic proteins and inhibiting the mitochondrial apoptotic pathway. Unlike Bcl-2 and Bcl-xL, Mcl-1 lacks a BH4 domain and contains an N-terminal PEST region with specific sequences for ubiquitination, phosphorylation, and caspase cleavage sites, which are important in regulating the half-life. Phosphorylation of at T92 and T163 by ERK inhibits Mcl-1 turnover, whereas phosphorylation by GSK3β at S159 facilitates proteosomal degradation. Increased expression of Mcl-1 is correlated with tumor survival and is associated with resistance of chemotherapeutics.

For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following: -Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol -100% ice-cold methanol only Fixation/permeabilization with Fixation buffer followed by 0.5 % Triton X-100 is not recommended due to dim signal.
