Product Description
HOMER1 is a scaffolding protein that is a major component of the post-synaptic density (PSD), a region in the plasma membrane of the post-synaptic neuron that interacts with the pre-synaptic active zone of upstream neurons. HOMER1 consists of two domains, a C-terminal coiled-coil domain that is required for HOMER1 dimerization, and an EVH1 domain involved in protein-protein interaction. Dimerized long-form HOMER1 (variants designated as Homer1b and Homer1c) is constitutively expressed in neurons, where it plays a role in neurotransmitter receptor signaling and trafficking by cross-linking associated proteins via the EVH1 domain. Truncated HOMER1 (Homer1a) is expressed as a result of neuronal activity, and lacking the coiled-coil domain, is incapable of dimerizing and cannot crosslink interacting proteins. Thus, Homer1a acts as a dominant-negative regulator of HOMER1 and is thought to provide negative feedback against excessive neuronal activity.
