Cytoplasmic actins, which belong to the microfilament system of cytoskeleton proteins, are some of the most conserved eukaryotic proteins expressed in mammals. α-smooth muscle actin is widely expressed in contractile cells, most notably the mural cells that line arteries and arterioles. Overexpression of α-smooth muscle actin is a hallmark of fibrosis, a thickening or scarring of damaged tissue.

Recommended Usage: Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. For flow cytometric staining using our True-Phos™ Perm Buffer, the suggested use of this reagent is ≤ 0.03 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application. * Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm. Additional Product Notes: For use in immunohistochemistry (IHC) on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend heat-induced antigen retrieval with either Tris-EDTA pH 9.0 or Citrate Buffer, 10X (Cat. No. 420902). For use in immunocytochemistry (ICC), we recommend fixation with Fixation Buffer (Cat. No. 420801) followed by permeabilization with 0.5% Triton-X. Previously Covance catalog# MMS-466S. For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with True-Phos™ Perm Buffer (Cat. No. 425401).
