PP2A alpha (also known as Serine/threonine-protein phosphatase 2A catalytic subunit alpha isoform or PPP2CA) is the catalytic subunit of protein phosphatase 2A (PP2A), which is involved in a wide range of cellular processes through its ability to dephosphorylate serine and threonine residues on target proteins. PPP2CA modulates the activity of proteins involved in cell cycle progression, such as cyclin-dependent kinases (CDKs), and is essential for the proper transition between different phases of the cell cycle, particularly G1/S and G2/M checkpoints. It also plays a critical role in DNA damage response, ensuring repair mechanisms are activated. Beyond cell cycle regulation, PP2A alpha influences various signaling cascades, including the MAPK, AKT, and WNT pathways, impacting cell proliferation, survival, and differentiation. Furthermore, PP2A alpha’s function in dephosphorylating tau proteins is significant in the context of neurodegenerative diseases, such as Alzheimer's disease, where abnormal phosphorylation of tau is a hallmark feature.

Recommended Usage: Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704). In human kidney tissue, this antibody is recommended for use at 10 µg/mL, as non-specific staining in tubules may be observed at lower concentrations. This product is not recommended for use in immunocytochemistry (ICC), intracellular flow cytometry (ICFC), or Western blot (WB).
