Zonula occludens-1 (ZO-1) is a scaffolding protein critical for the assembly and maintenance of tight junctions in epithelial and endothelial cells. It links transmembrane tight junction proteins such as claudins and occludin to the actin cytoskeleton, thereby maintaining cell polarity and barrier integrity. ZO-1 also participates in signal transduction pathways that regulate cell proliferation and differentiation. Loss or mislocalization of ZO-1 is associated with impaired barrier function and has been implicated in various pathological conditions, including cancer and inflammatory diseases. Recommended Usage: Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704). For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following: -Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100 -Fixation buffer (Cat.
