Product Description

SWAP70 was initially isolated as a component of an isotype switch recombination complex (SWAP) from activated B lymphocytes. It translocates from cytoplasm to the nucleus upon B cell activation. SWAP70 interacts with F-actin and RAC, binds to PI(3,4)P 2 , and regulates membrane ruffling and lamellipodia formation in response to growth factor stimulation.

Recommended Usage: Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1-0.5 µg per ml. For immunoprecipitation, the suggested use of this reagent is 10-30 μg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Structure: Contains a PH domain, which is required for binding of phosphatidylinositol 3,4,5-trisphosphate, a coil-coil domain, and a domain weakly homologous to DH domains. Theoretical molecular weight is 69 kD.
Distribution: SWAP70 is localized to cytoplasm in resting B-cells, recruited to plasma membrane, and translocated to the nucleus upon B-cell activation.
Function: SWAP70 is a guanine nucleotide exchange factor (GEF) which binds phosphatidylinositol trisphosphate and transduces signals from tyrosine kinase receptors to RAC. SWAP70 is required for membrane ruffling and lamellipodia formation during cytoskeletal F-actin rearrangement.
Interaction: Interacts with cytoskeletal F-actin and Rac to regulate actin remodeling. Associates with NPM1, NCL, and PARP1 in switch-associated protein complex.
