Desmin is an intermediate filament protein found primarily in muscle cells, where it plays a critical role in maintaining the structural integrity and organization of the cell. Desmin forms a network that connects myofibrils to the sarcolemma and cytoskeleton, providing mechanical support during muscle contraction. This ensures proper alignment and function of the sarcomere, the basic contractile unit of muscle. Desmin also plays a role in muscle mitochondrial dynamics, as loss of Desmin results in defects in mitochondrial morphology, location, and function.

Recommended Usage: Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704). For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following: Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100 Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol 100% ice-cold methanol only For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803), True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401). Signal is brightest using True-Nuclear™ Transcription Factor Buffer Set.
