Sestrin 2 (SESN2) is part of the sestrin family of evolutionally conserved stress inducible proteins. SESN2 plays a role in cell growth and survival by protecting cells against oxidative and genotoxic stress. SESN2 reduces oxidative stress by activating Nrf2 and inhibits mTOR activation through AMPK. SESN2 is an intracellular leucine sensor which negatively regulates protein translation in response to ER stress by interacting with the TORC1 signaling pathway through GATOR2. SESN1 is a paralog of SESN2. Diseases associated with SESN2 include nodular ganglioneuroblastoma and pulmonary emphysema.
