Gasdermin D (N-terminus) is a member of the Gasdermin family of pore-forming proteins involved in pyroptosis. Gasdermin-D is vital to inducing pyroptotic cell death in response to pathogen and damage-associated molecular patterns, thus performing a role crucial to innate immune defense. Gasdermin-D is localized to the cytosol in an inactive state but, following cleavage into N and C-terminal fragments by inflammatory caspases Caspase-1, 4, or 5, the N-terminal fragment translocates to the plasma membrane where it forms large oligomeric pores. Pore-formation results in a disruption in the cell’s osmotic potential and subsequent cell lysis. Secretion of pro-inflammatory cytokines IL-1β and IL-18 from bone marrow dendritic cells (BMDCs), bone marrow derived macrophages (BMDMs), and neutrophils is also dependent upon Gasdermin-D, both through release by cell lysis and by direct passage through pores formed by its N-terminal fragment in the plasma membrane. Gasdermin-D exhibits bactericidal properties through the binding of its N-terminus to the cardiolipin of bacterial cell membranes and subsequent membrane perforation.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting and immunoprecipitation the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: This clone was tested for ICC using NTERA-2 (negative control) and PC-3 (positive control) cells fixed with 4% paraformaldehyde and permeabilized with either methanol or Triton X-100. Both methods resulted in weak staining with high background. W18029B is predicted to recognize the pro-pyroptotic N-terminal fragment of cleaved Gasdermin-D. The immunogen used to generate this clone is fully contained within the sequence of the N-terminal fragment.
