Background Transportin-1 (TNPO1), also known as MIP or karyopherin-β2, is a nuclear import protein that binds to nuclear localization signals (NLSs) on various proteins to help shuttle them from the cytoplasm to the nucleus (1-3). While initially discovered to shuttle hnRNP proteins, transportin-1 has also been shown to import other important regulatory proteins, such as c-Fos and ADAR1 (1,4,5). Transportin-1 plays a role in HIV replication, as it imports and helps uncap the capsid protein (6). Transportin-1 has been implicated in familial ALS, due to its impaired binding affinity to mutations in the FUS-NLS region. Reduction in transportin-1 binding correlated with the time of ALS disease progression in these mutants (7). Transportin-1 can also accumulate in FUS inclusions in wild-type FUS ALS (8). Additionally, transportin-1 plays a role in circadian rhythms by importing PER1 to the nucleus (9).
