Product Usage Information Western Blotting: 1:1000 Immunohistochemistry (Paraffin): 1:50 - 1:200 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Background Palmitoyl-protein thioesterase 1 (PPT1) is a depalmitoylation enzyme that removes palmitate groups from S-acylated proteins (1). PPT1 localization has been demonstrated in multiple compartments, including membrane rafts, the nucleus, lysosomes, the Golgi apparatus, synaptic vesicles, axons, and the endoplasmic reticulum (ER) (1,2). Mutations of the PPT1 protein can cause neuronal ceroid lipofuscinosis (NCL) type 1 (CLN1), a lysosomal storage disorder characterized by brain atrophy, mental and motor retardation, seizures, and retinal degeneration, ultimately resulting in blindness (2,3). In healthy, aged brains, PPT1 and other CLN proteins have been identified as lipofuscin constituents (4).
