Product details:
Human SDF1 ELISA Kit ab100637 is a sandwich ELISA to measure Human SDF1 in plasma, cell culture supernatant with a sensitivity of 42 pg/ml.
How the assay works
This assay employs an antibody specific for Human SDF1 coated on a 96-well plate. Standards and samples are pipetted into the wells and SDF1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human SDF1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SDF1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

1. Add standard or sample to each well used. Incubate at room temperature
2. Wash and add prepared biotinylated antibody to each well. Incubate at room temperature.
3. Wash and add prepared streptavidin solution. Incubate at room temperature.
4. Add TMB Development Solution to each well. Incubate at room temperature.
5. Add Stop Solution to each well. Read at 450nm immediately.
Optimization may be required with urine samples.

SDF1 beta and SDF1 alpha also known as CXCL12 are small chemokines playing important roles in several cellular processes. These signaling proteins have a molecular mass ranging from approximately 7 to 15 kDa depending on post-translational modifications. They are secreted proteins expressed in various tissues including bone marrow heart liver and brain. SDF1 beta and SDF1 alpha function by binding to the chemokine receptor CXCR4 mediating cellular responses through signal transduction mechanisms.
Biological function summary
SDF1 beta and SDF1 alpha regulate hematopoietic cell trafficking and homing. These proteins are significant for directing migration of immune cells and stem cells by forming part of a ligand-receptor complex with CXCR4. This interaction influences cell survival proliferation and inflammation. SDF1 also participates in organ development and promotes angiogenesis during tissue repair.

SDF1 beta and SDF1 alpha are central to the CXCL12-CXCR4 axis important in signaling pathways like G-protein coupled receptor signaling and JAK-STAT signaling. These pathways contribute to immune response modulation and stem cell maintenance. Notably the interaction with CXCR4 is essential in the SDF-CXCR4 axis and effects are also mediated with other proteins like CXCR7 further impacting cellular signaling.
SDF1 beta and SDF1 alpha have associations with cancer metastasis and HIV infection. The SDF1-CXCR4 interaction represents an important point in metastatic spread since it aids tumor cell migration and invasion. This chemokine binding also serves as an essential co-receptor for HIV entry into cells implicating it in disease progression. Alterations in the SDF1-CXCR4 axis connect to oncogenic processes and viral infections alongside proteins such as CCR5 contributing to HIV pathway interactions.
