CA150 also known as TCERG1 (transcription elongation regulator 1) is a nuclear protein with a molecular mass of approximately 150 kDa. This protein localizes mainly in the nucleus where it is expressed in various cell types. Mechanically CA150 interacts with RNA polymerase II the enzyme responsible for transcribing DNA into messenger RNA. It regulates transcription elongation the phase where RNA polymerase II transcribes DNA into longer RNA strands. CA150 acts by modulating the phosphorylation of RNA polymerase II which is essential for progressing the transcription process from initiation to elongation.
Biological function summary
The transcription elongation regulator 1 protein modulates the expression of genes by being part of transcriptional complexes. CA150 has important interactions with the pre-mRNA splicing machinery impacting how precursor mRNAs are processed into mature mRNAs. This includes altering splicing patterns and affecting alternative splicing a process required for generating protein diversity. The protein influences cell cycle progression and cellular responses to DNA damage highlighting its important role in gene expression regulation and cellular homeostasis.
Pathways
Various genes and cellular processes show links to CA150's function. This protein is part of the transcriptional regulation pathway and is involved in the response to DNA damage signaling pathways. In these pathways CA150 directly interacts with proteins like cyclin-dependent kinases (CDKs) and the splicing factor SF3b to coordinate transcription and splicing processes as DNA is reproduced and repaired. Its ability to connect transcription with RNA processing makes CA150 integral to regulating gene expression at multiple stages.
Transcription elongation processes involving CA150 play a role in aberrant splicing events. Alterations in CA150 function are implicated in neurological disorders such as spinal muscular atrophy (SMA) where splicing defects occur. Moreover CA150 links with proteins like survival motor neuron (SMN) protein in these pathways. It also demonstrates connections to cancer where dysregulated gene expression and splicing defects can lead to tumorigenesis. Here modifying CA150 activity could influence oncogenic pathways suggesting that it might have potential as a therapeutic target.
