The Iron Regulatory Protein 2 (IREB2) also known as Iron Responsive Element Binding Protein 2 (IRP2) is an essential regulator of cellular iron homeostasis. IREB2 mechanically binds to iron-responsive elements (IREs) on mRNA controlling the post-transcriptional expression of target genes involved in iron metabolism. The protein has a molecular weight of approximately 105 kDa and is expressed in various tissues including the liver brain and pancreas. Its regulation is strongly controlled by iron availability with high iron levels leading to its degradation.
Biological function summary
IREB2 regulates the stability and translation of mRNAs encoding proteins linked to iron uptake storage and utilization. It plays an important role outside of a complex acting primarily on its own to bind transcripts that include ferritin and transferrin receptor mRNAs. Its activity increases under conditions of low iron availability promoting iron uptake and reducing iron sequestration to meet metabolic needs. Through these mechanisms IREB2 helps maintain iron supply for essential enzymes and cellular processes.

IREB2 acts as a central player in the iron metabolism pathway influencing both iron import and storage. The pathways it participates in are essential for maintaining cellular and systemic iron balance. It works in coordination with IRP1 forming a regulatory network that adjusts iron homeostasis. Under varying iron concentrations IREB2 modulates expression of its target genes involved in iron transport and storage within the framework of the iron-sulfur cluster biosynthesis pathway.
Aberrant IREB2 activity links to neurodegenerative diseases such as Alzheimer's disease where iron dysregulation is often observed.
