ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Platelet Glycoprotein IIb+IIIa concentrations in Human platelet-rich plasma and platelets.
A Platelet Glycoprotein llb + llla specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Platelet Glycoprotein llb+llla specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Platelet Glycoprotein llb + llla captured in plate.
The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Platelet glycoprotein IIb/IIIa also known as GP IIb/IIIa GP IIb/IIIa complex or integrin αIIbβ3 is an important protein integrin present on the surface of platelets. It has a molecular mass of approximately 130 kDa for each subunit and is expressed mainly in platelets and megakaryocytes. The protein functions as a receptor complex essential for platelet aggregation. Upon activation it changes conformation enabling it to bind fibrinogen and other adhesion molecules facilitating the cross-linking of platelets and forming a platelet plug.
Biological function summary
Platelet glycoprotein IIb/IIIa mediates the final common pathway for platelet aggregation by forming part of the integrin superfamily complex acting as a bridge between adjacent platelets. It converts mechanical stimuli into chemical signals important for thrombus formation. Interactions with another key platelet glycoprotein GP Ib-IX-V strengthen the adhesion and aggregation process ensuring an efficient hemostatic response to vascular injury.
Pathways
Platelet glycoprotein IIb/IIIa plays a significant role in the hemostasis and thrombosis pathways important for maintaining blood flow control. The protein is involved in mediating the platelet activation pathway with interactions notably including the von Willebrand factor and fibrinogen. This integrin bridges the interaction between these pathways which also involves associated molecules such as thrombin and thromboxane A2 playing a comprehensive role in clot formation.
Platelet glycoprotein IIb/IIIa is directly implicated in conditions such as acute coronary syndromes and stroke. Abnormalities or overactivity in this integrin can lead to excessive platelet aggregation and thrombosis which are pathological states in these diseases. Therapeutic antibodies targeting GP IIb/IIIa such as abciximab and other antagonists are often used to reduce risks associated with these disorders by preventing excessive clot formation. Additionally it relates closely to GP Ib-IX-V in pathological contexts influencing the development of thrombotic disorders.
