Adenine Nucleotide Translocase (ANT) also known as ADP/ATP translocase is a mitochondrial membrane protein with a mass of around 30 kDa. It facilitates the exchange of adenosine diphosphate (ADP) and adenosine triphosphate (ATP) across the inner mitochondrial membrane playing an important role in cellular energy metabolism. ANT is highly expressed in tissues with significant energy demands such as muscle and heart muscle where efficient ATP production and transport are critical.
Biological function summary
ANT functions as a part of the mitochondrial permeability transition pore (mPTP) complex where it contributes to the regulation of mitochondrial membrane potential and cell apoptosis. By mediating the flow of nucleotides like ATP and ADP ANT ensures proper cellular respiration and energy balance. Its presence in the mPTP complex links ANT to mitochondrial responses during cellular stress conditions influencing cell survival and death.
Pathways
ANT is integrally involved in the oxidative phosphorylation pathway a process essential for ATP synthesis in mitochondria. ANT cooperates closely with ATP synthase facilitating the movement of nucleotides that support ATP generation. Its role connects with other proteins such as cytochrome c oxidase which plays a part in the electron transport chain highlighting ANT's importance in bioenergetics and cellular respiration pathways.
ANT has a connection to mitochondrial myopathies and cardiac ischemia due to its central role in energy metabolism. Abnormal ANT function can lead to impairments in ATP transport contributing to muscle weakness in myopathies and energy deficits in heart tissues during ischemia. ANT also interacts with proteins such as cyclophilin D which modulate the mPTP complex linking ANT to conditions involving mitochondrial dysfunction.
