FBXL17 also known as F-box/LRR-repeat protein 17 functions as a member of the F-box protein family. These proteins form part of the SCF (SKP1-CUL1-F-box protein) complex a type of E3 ubiquitin ligase involved in protein degradation. FBXL17 has a molecular mass of approximately 49 kDa. It is expressed in various tissues with notable presence in the liver lung and kidney. By interacting with SCF FBXL17 plays a role in targeting specific substrates for ubiquitination which can regulate their stability and function.
Biological function summary
FBXL17 influences diverse processes through its role in protein ubiquitination. As a component of the SCF complex it regulates cell cycle progression and cell signaling by mediating the degradation of proteins involved in these pathways. Through protein interaction FBXL17 modulates cellular responses and assists in maintaining cellular homeostasis. It does this by recognizing and binding to phosphodegrons on substrates therefore determining their ubiquitination.
Pathways
FBXL17 interfaces with the cell cycle and signal transduction pathways. It is involved in regulating proteins like cyclins and cyclin-dependent kinases which are fundamental for cell cycle control. Additionally FBXL17 participates in the Wnt/β-catenin signaling pathway. By influencing the stability of proteins like β-catenin FBXL17 can affect gene expression and cellular activities related to these pathways.
FBXL17's dysregulation or altered expression may have connections to cancer and neurodegenerative diseases. In cancer FBXL17 can influence tumorigenesis by altering the degradation of oncoproteins and tumor suppressor genes. Its role in regulating the Wnt/β-catenin pathway associates it with disorders like colorectal cancer. FBXL17's interactions with other ubiquitin ligases such as FBXW7 may contribute to understanding its precise role in disease pathology.
