Dub3 also known as Deubiquitinase 3 is an enzyme that acts as a deubiquitinating enzyme (DUB). This enzyme removes ubiquitin from proteins an important process in regulating protein stability and function. Dub3 has a molecular mass of approximately 59 kDa and is expressed in various tissues including high expression in the uterus placenta and testis. Its activity is important for maintaining proper levels of cellular proteins by reversing the ubiquitination process.
Biological function summary
Deubiquitinase 3 plays a significant role in regulating cell cycle progression and DNA damage response. It is often found within a larger complex where it interacts with other proteins to exert its function. This enzyme modulates the activity of several key proteins such as Cyclin A and Cdc25A by controlling their ubiquitination status which influences cell division and repair mechanisms in the cell. By maintaining these processes Dub3 ensures the fidelity of the cell cycle and genomic integrity.
Pathways
The function of Dub3 integrates into the ubiquitin-proteasome system and the DNA damage repair pathway. In the ubiquitin-proteasome system Dub3 removes ubiquitin chains from target proteins which determines their fate either for degradation or stabilization. On the DNA damage repair front Dub3 contributes to pathways involving proteins like p53 by deubiquitinating and stabilizing them allowing for an adequate response to DNA damage signals and maintaining cellular homeostasis.
Dub3 has been linked to certain cancers such as breast cancer and prostate cancer. In these cases its dysregulation leads to abnormal protein stabilization that can promote tumorigenesis. Dub3 affects oncogenic pathways involving proteins such as MYC and p53 which show altered levels and activity in cancer. Understanding the role of Dub3 in these disorders enables researchers to consider it a potential therapeutic target providing insights into how its modulation might benefit treatment strategies.
