Dyrk1B also referred to as Mirk is a protein kinase with a molecular mass of approximately 65 kDa. It belongs to the DYRK family which shares characteristics with dual-specificity kinases. Dyrk1B plays a role in phosphorylation activities which involves the addition of phosphate groups to target proteins altering their activity. This kinase is expressed in various tissues notably in muscle and certain cancer cells where it influences cellular processes such as proliferation and survival.
Biological function summary
Dyrk1B affects cell cycle regulation and differentiation. It is involved in promoting cell cycle exit and muscle differentiation by phosphorylating specific transcription factors. Dyrk1B exists as a monomer not requiring interaction with other proteins to function. By regulating transcription factors it contributes to the control of growth-restrictive environments and supports cell survival under stress conditions.
Pathways
Dyrk1B interacts with multiple signaling pathways including the Ras/MAPK and PI3K/Akt pathways. In these pathways Dyrk1B interacts with proteins such as p27Kip1 and FOXO impacting cellular quiescence and survival. Its influence in these pathways suggests a role in controlling important mechanism of cell cycle arrest and resistance to stress.
Dyrk1B has relevance to certain cancers and metabolic disorders like type 2 diabetes. In pancreatic cancer Dyrk1B supports cell survival and resistance to chemotherapy. Additionally its dysregulation impacts insulin resistance in metabolic disorders. Cancers often show upregulation of Dyrk1B working in conjunction with kinases like AKT to enhance tumor survival and progression providing potential targets for therapeutic intervention.
