PASK also known as PAS domain-containing serine/threonine-protein kinase is a protein with a mass of approximately 142 kDa. This kinase expresses in various tissues including the liver heart and skeletal muscle. PASK contains PAS (Per-ARNT-Sim) domains that allow adaptation to environmental and cellular signals. The protein kinase plays an important role in regulating energy balance and metabolism through phosphorylation of downstream targets.
Biological function summary
PASK functions as a metabolic sensor that links changes in cellular energy levels to the regulation of gene expression and metabolic processes. It does not form part of a well-characterized complex but interacts with other proteins to exert its effects. PASK influences insulin secretion in pancreatic beta-cells and regulates glucose homeostasis. It plays a role in lipid metabolism by modulating the activity of various metabolic enzymes and transcription factors.
Pathways
PASK participates in the insulin signaling pathway and nutrient-sensitive pathways influencing energy homeostasis and cellular metabolism. It interacts closely with proteins such as AMP-activated protein kinase (AMPK) and mTOR which are themselves central regulators of cellular energy balance. These interactions place PASK as a node in the complex network controlling energy storage and expenditure.
PASK has been linked to metabolic diseases such as type 2 diabetes and obesity. Altered expression or mutations in PASK can disrupt metabolic regulation leading to insulin resistance. Additionally studies have shown connections between PASK and cardiovascular diseases due to its involvement in lipid metabolism and glucose homeostasis. It interacts with proteins like peroxisome proliferator-activated receptor gamma (PPARγ) which plays a role in fat storage and glucose metabolism highlighting PASK's importance in these conditions.
