LEFTY2 also known as Left-Right Determination Factor 2 is a member of the transforming growth factor beta (TGF-β) superfamily. This protein has a molecular mass of approximately 38 kDa. LEFTY2 is expressed mainly in visceral endoderm and left side mesoderm tissues. The gene encoding LEFTY2 is found within the human chromosome 1. This protein plays a mechanical role in the regulation of cellular signaling and developmental processes.
Biological function summary
LEFTY2 functions as an important regulator in ensuring proper left-right asymmetry during embryonic development. It is a monomeric protein that is not part of a larger complex. LEFTY2 contributes to the modulation of Nodal signaling inhibiting excessive activity that can disrupt normal development. This modulation ensures the establishment of asymmetric organ positioning and plays an important role in maintaining developmental balance in the body.
Pathways
LEFTY2 integrates into the Nodal signaling pathway and the Activin signaling pathway. The interaction with Nodal proteins narrows down Nodal signaling ensuring the accurate development of the left-right axis. LEFTY2 also relates functionally and antagonistically to other proteins like Nodal and Activin A balancing their signaling actions to regulate developmental processes.
LEFTY2 has a connection with asymmetry-related conditions such as situs inversus where organ placement within the body is mirrored from normal positions. Additionally research links irregular LEFTY2 expression to some cancer types including ovarian cancer. Through these disorders LEFTY2 interacts with proteins like Nodal which plays a role in cancer cell proliferation by influencing the signaling pathways that govern cellular growth and differentiation.
