FBXO9 also known as F-box only protein 9 functions as part of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex. This complex tags proteins for degradation by attaching ubiquitin molecules. FBXO9 specifically acts as a substrate recognition component targeting proteins for ubiquitination and subsequent proteasomal degradation. The mass of FBXO9 is around 47 kDa. It is expressed in various tissues with a significant presence in the lung and spleen.
Biological function summary
FBXO9 participates in the regulation of protein stability and turnover. As a component of the SCF complex it plays a role in controlling cell cycle progression and signal transduction. Its function in recognizing and binding specific proteins identifies them for ubiquitin-mediated degradation regulating protein levels within cells. This process is important for maintaining cellular homeostasis and responding to changes in the cellular environment.
Pathways
FBXO9 interacts with processes involved in protein degradation and cell cycle regulation. It is a part of the ubiquitin-proteasome pathway important for removing damaged or misfolded proteins. In addition FBXO9 influences the cell cycle pathway by targeting regulatory proteins for degradation affecting their availability and function. It works alongside proteins such as SKP1 and cullin-1 in these pathways ensuring precise control over cell cycle progression.
FBXO9 has links to certain types of cancers and neurodegenerative diseases. Aberrations in its function may lead to disruptions in protein homeostasis contributing to tumorigenesis through unchecked proliferation due to altered cell cycle regulation. In neurodegenerative diseases FBXO9's role in protein degradation becomes significant as accumulation of malfunctioning proteins can lead to cell death. Interactions with proteins like p27 a cyclin-dependent kinase inhibitor highlight its importance in these disease contexts.
