The eNOS iNOS and nNOS commonly called endothelial inducible and neuronal nitric oxide synthase respectively play an important role in the production of nitric oxide (NO) from L-arginine. eNOS is typically expressed in endothelial cells iNOS in immune system cells and nNOS in neurons. eNOS has a molecular mass of approximately 133 kDa while iNOS is around 131 kDa and nNOS is about 160 kDa. These enzymes are mainly located in vascular tissues macrophages and nerve tissues. They exhibit distinct expression patterns depending on their cellular environments reflecting their specialized roles in different tissues.
Biological function summary
Nitric oxide synthases produce NO an important signaling molecule that affects vasodilation neurotransmission and microbial killing. eNOS iNOS and nNOS all produce NO but differ in their regulation and temporal expression. eNOS functions constitutively and is calcium-calmodulin dependent. In contrast iNOS is calcium-independent and gets induced under inflammatory conditions. nNOS also operates constitutively and modulates neuronal communication. While these enzymes do not form a multimeric complex themselves they often associate with other proteins to exert their functions effectively.

Nitric oxide synthases significantly contribute to the nitric oxide signaling pathway. In this context eNOS is pivotal in the regulation of blood pressure and flow by modulating vascular tone. It interacts with calmodulin and other enzymes like phosphoinositide 3-kinase (PI3K) in endothelial cells. nNOS in the nervous system participates in synaptic plasticity and interacts with proteins such as NMDA receptors to influence learning and memory pathways. iNOS meanwhile becomes highly active in immune response pathways particularly involving macrophages where NO acts as a defense mechanism against pathogens.
Abnormal nNOS activity links to neurodegenerative conditions like Alzheimer's disease. nNOS through excessive NO production can lead to neuronal damage. In cardiovascular disorders eNOS dysfunction associates with hypertension and atherosclerosis due to impaired NO-mediated vasodilation. Here eNOS's interaction with proteins like caveolin-1 may alter its functionality.
